Schizophrenia is associated with a widespread loss of synaptic connections in the brain. This loss follows a surprising, organized pattern. Researchers used specialized positron emission tomography (PET) imaging to examine these connections in living human brains.
The study involved 122 individuals, including 29 diagnosed with schizophrenia. People with schizophrenia showed a significant reduction in synaptic connections across various brain regions. These areas included frontal and temporal lobes, as well as regions involved in memory and emotion. The left side of the brain experienced considerably greater loss than the right side.
This synaptic pattern did not align with brain volume changes observed in standard magnetic resonance imaging (MRI) scans. This suggests that synaptic loss and changes in brain volume may represent distinct biological processes. The brain regions with the most significant synaptic losses also had high concentrations of receptors for key neurotransmitters. These include serotonin, gamma-aminobutyric acid, and glutamate.
This finding suggests that the molecular characteristics of specific brain regions may influence their vulnerability to changes related to schizophrenia. Computer simulations based on the brain's structural connections identified a region in the left frontal lobe. This area may serve as a starting point from which synaptic loss could spread to connected regions.
These findings indicate that synaptic loss in schizophrenia is not random. It follows the brain's molecular and connectivity architecture. Understanding this pattern could help identify intervention points for preserving or restoring brain function. Future research will explore how synaptic loss progresses over time and responds to treatments. This could lead to more precise and personalized approaches to schizophrenia care.
